ABSTRACT: The problem of the mechanism of action of the oubatances mon-
tioned in the title is still insufficiently investigated. It
is only evident that at least 2 factors are important in this
mechanism: a) the capacity of anesthetics to be adsorbed on
the cell surface, and b) their interaction (chemically speak-
ing) with structures of the receptor. Many investigations
have already been carried out concerning the relation betwe~~_n
chemical structure and biological effect of anestheticF. It
became apparent that the anesthetizing activity of a compound
depends on the presence of an aromatic acid in its molecule.
This acid is linked to the nitrogen-containing main group by
means of an enter binding or a binding which is isosteric t)
Card 1/3 the eater binding. The interdependence of the structure and
On the Relation Between the Anesthetizing Properties SOV/20-126-6-58/67
of Anesthetics and Their Surface Activity
the first phase of the biological effect, i.e. the penetration
of the substanceto the place of reaction and its ae,orption on
the cell surface, has, in spite of its importance, not yet
been determined. A survey on literature is given ~Refs 1-9,
11,14,16), in which the contradictory results of individual
authors are noticed. Thus the question of the interdependence
of the anesthetizing effect and the surface propertiec of
anesthetics cannot be regarded as completely solved. The
present investigation presents a further comparison between
these 2 factors in the case of various anesthetics. The termi-
nal anesthesia was studied oi the cornea of a rabbit according
to the Renlye--Valet method New compounds were investigated:
Ksilokain (xylocaine), Me,!zkain (mezocaine - deri-~atives of
di- and trimethyl azilides), Oksikain (oxycaine) Nr 4 and
Nr 12 (ester derivatives of the paraoxy benzoic acid,;),as
well as novocaine, cocaine, dicaine, and sovcaine. It was
proved that oxycaines, sovcaine, dicaine, mesucaine, and xylo-
cAine have a high surface activity. Their order in regard of
the decroat3ing abovo mentioned activity is the one Iriven
Card 2/3 above. They are followed by cocaine and novocaine ('
Fig 1).
On the Relation Between the Anesthetizing kropertiea SOV/20-126-6-58/67
of Anesthetics and Their Surface Activity
There is a cymbathic dependence between their capacity of
effecting a terminal anesthesia and the surface activity of
the said substances (Tablel, Fig 2). There exists a parallelism
between these two indexes. With the increase of the pH from
5.0 to 7.0 the effect of all anesthetics on the surface ten-
sion increases in which case the order of substances remains
unchanged at these two pH values. The parallelism is not always
very mark-~ but results point to a direct relation between
the said properties. They are higher in free bases than in the
case of anesthetics in cation form. There are 2 figures, 1
table, and 16 references, 2 of which are Soviet.

ABSTRACTs The purpose of this paper is the detection of a relation between
the state (shape) of protein macromolecules in a solution and
their power of solubilizing substances of low solubility inwater
(especially hydrocarbons). Gelatin was used as protein componento
Before carrying out the experiments, the gelatin was cleaned by
means of high-voltage many-chamber electrodialysis according to
a method developed by V. A. Kargin and T.A. Matveyeva(Ref 11).
Benzene was used as dissolved substance. The quantitative
measurement of the solubilization was carried out according to
the refractometer method of A. 1. Yurzhenko (Ref 19). The
presence of drops of the emulsified excess substance did not
influence the experimental results. The refraction indices were
measured by means of refractometers of the Abbe and Pulfrich

solution mainly depends on the PH value of the substance.
Therefore, this quantity was investigated first of all. The
PH value of the substance was varied by addition of HCI or NaOH
within the interval from p112 to PH 11, The iooeloctric point of
the dialyzed gelatin is PH 5.2. The measuring of the
solubilization of benzene is discussed in short. After the
corresponding calculations the dissolubility of benzene in
gelatin solutions of various concentrations was found, and the
results of these measurements at 200 are given in a table. For
any investigated concentrationt the highest disoolubility of
benzene was observed at the isoelectric point. In the acid and
in the alkaline region, solubility is noticeably lower than in
the isoelectric point, but nevertheless it is higher than in
pure water. The maximum can be explained by coagulation of the
furcated chains of the gelatin chains in the isoelectric point.
In connection with the above considerations, it was interesting
to investigate the variation of the viscosity of gelatin
solutions after the dissolution of benzene in them. This
viscosity was measured by means of ar. elastoviscosimeter.

the substance (at a temperature of 200). Por PH 5.2, the
ructural viscosity in the isoelectric point is slightly
t
ioreased after solubilization. A possible explanation of this
/'henomenon is discussed in short. In the acid and in the alka]4.ne
region, structural viscosity sharply decreased after the
dinnolution of bonzone. Tho third diagram ahowa tho dopondenco
of the solubilization of benzene on the gelatin concentrat 'ion in
the solution. UP to 0-5~ of gelatin content, there is a linear
increase in solubilzation, but at higher concentrations the
solubilization of benzene becomes constant. At 300, there is np
aharacteriatic maximum of oolubilization in tho isooloctric
point, but nevertheless the solubility of benzene in gelatin
solution is somewhat higher than in water. There are 3 figures
and 17 references, 13 of which are Soviet.
