TITLE: Synthesis of 6-Amino-7-Methillptirine Derivatives as Possible Antimetabolites PERIODICAL% Zhurnal*obshchey khimii, 1960, Vol 30, Nr 1, PP 327- 332 (USSR) ABSTRAM Several derivatives of 7-methylpurine were synthesized for future study of their antimetabolic action. The following compounds were synthesized and described: 2.6-dichloro-7-methylpurine M, 7-methyladenine (II), 2-chloro-6-ethyleneimino-7-methylpuriiie (III), 2-chloro- 6-monoethanolamino-7-meth,jlpurine (IV), 2-chloro-6- diethanolamino-7-methylpLirine (V) , 6-diethylamino-7- met 1purine (VI), 6-motioei;liailolamino-7-m(-,thylptirine (VI~I, eth I ester of N-(2-chloi-0-7-lnc,'thylptirine-6-Lrly- cine (VIM-, ethyl ester of N-(2-cfiloro-7-methylpurine- 6)-dl- CL -alanine IX), and ethyl ester of N-(7-methyl- purine-6)-glycine R). Compound I was prepared by the Card 1/4 modified method of J. Davell for preparation of Synthesis of 6-Amino-7-Methylpurine Deriva- 77411 tives as Possible Antimetabolites SOVI/79-30-1-72/78 Llrichloropurine (J. Am. Chem. Soc., 73, 2937 (1951)): freshly distilled phosphoryl chloride was boiled with theobromine and dimethylaniline; the residue left after distillation of P001 3 was neutralized with Na 2co3 and the precipitate washed with 2% KOH and recrystallized from water (yield 25%; mP 197.5-1960). Compound III waa made by letting the mixture of ethyleneimine, 2,6- dichloro-7-methylpurine, and 1% NaOH stand for 18-20 hr at room temperature. 2-Chloro-6-aminosubstituted 7- methylpitrines kcompounds IV, V, VIII, a6d IX; see Table A) were synthesized by boiling the alcohol solu- tion of compound I with the corresponding amine. The 6-aminosubstituted 7-methylpurines (compounds II, VI, and X) were obtained by the modified method of E. Fischer (Ber., 30, 21100 (1897); ibid, 31, 1011 (1898)), i.e., by boiling 2-ch1oro-6-amLnosubqtituted '(-methyl- purine with HI (d 1.50) and red phosphorus. Compound VII was prepared by reacting ethylene oxide with a solution of 7-methyladenine in 25% acetic acid. Biological aotion of compounds III, V, VII, II, VI, and X was studied in the laboratory of experimental chemotherapy. In puppies, the described disorders were less ked and recovery was more rapid. Thus, the conserved central tracts were able to take care of the functions affected by the operation.
