pressure or sudden lowering of circulating volume was studied in 10 healthy sub-
jects after Lv. injection of hypertonic saline (20 ml. 15% NaCl) with Jeffers'

Coproporphyrins were extracted from the acidified urine with ethyl acetate. The
extracts were washed and the ~oproporphyrins transferred into diluted RCI and
determined by colorimetry in the Beckmann photometer at 4010 A. For semi.-
quantitative determination a simple fluorometric method may be sufficient: To
2 ml. of urine are added 0.4 ml. of acetate buffer (4 parts of glacial. acetic acid +
I part of saturated Na acetate) and the mixture shaken with 10 ml. of ether. To
5 ml. of the ether phase are added 3-5 drops of 0.116 solution of iodine. After 10
min. 0.25 ml. of 2N HC1 and after 10 min. 4.75 ml. of watee tire udded with shakIng.
The Intensity of the fluorescence is compared with standard solutions of copro-
porphyrin in HCL As a source of UV-11ght a mercury vapour lamp with a fluid

The total coproporphyrin (1) in fresh urine in lead poisoning is composed of
1-12 O/o preformed I and 38-99 O/o I-precursor(s). This is the highest value for
I rcursor so far reported. No evidence of an incrcucd excretion of preform-
ell in lead poisonin CoUld be obtained; the increased urinary level is due to
increased ex( f precursor(s). After rapid voiding of urine the precursor
is quickly converted to I in the dark in presence of oxygen. On exposure to
light there is rapid breakdown to non-porphyrin substances. In urine in a
test tube exposed to diffuse daylight, the total I content dropped to a small
percentage of the original value after only 2 hr. Samples for analysis must
therefore not be exposcil to ligh4 even for short periods.

Abstract : A study was madet of the coproporphIrrin precursor lcmal in
the urine of healthy subjects and of those sv ffering from
lead poisoning. Methyl ethers of I :?orphyrin 4ere extracted
from the urine by various methods. Coproporoyrin vas se-
parated by colwm chromatography and its isiomers)'I and II,
by paper chromatography. It vas found that in lemlpoiso-
ning the urine contained predominantly a preeursor of co-
proporphyrin III and only a small amount of coproporphyrin.

studied in experimezLts on mice and in observations on
50 patients. Edema, appeared in 6 out of 50 patients.
The use of these drugs is not recommended in diseases
accompanied by edema.. Their antidiuretic effect is
attributed to the stinulation of the tubular reabsorp-
tion of water.
